Quantitation of lipids by charring on thin-layer plates and scintillation quenching: application to ceramide determination.

چکیده
A method is described for the quantitative determination of lipids after separation by thinlayer chromatography: the lipid spots are visualized with a charring spray and the darkness of the spots is determined by measuring the quenching produced when the powder is suspended in a scintillation counting gel containing a radioactive sample. A standard curve produced with 2 to 20 pg of ceramide was Iinear, so only two standard spots (in duplicate) are needed on each thin-layer plate. Using the vertical sample streak technique, one can analyze three unknown samples (in duplicate) on each plate. This technique allows one to use the quantitative thin-layer approach without purchasing a special apparatus for the purpose. As an example of the method, we analyzed tissues for ceramide content, after first removing much polar lipid by a liquid/liquid partitioning technique. This is the most convenient method available for determining this minor, but widely distributed lipid.

Determination of glucocerebroside, sphingomyelin, free fatty acid and total lipid by thin-layer chromatography and charring-scintillation quenching.
A previously described method has been extended to various specific lipids of liver and brain. The basic method involves thin-layer chromatography followed by charring to reveal the bands. The intensity of each band is determined by suspending the silica gel in a radioactive scintillation gel and measuring the optically quenched activities. The lipids are extracted with hexane/isopropanol and, ...

THIN-LAYER CHROMATOGRAPHY (TLC) has been widely applied to the qualitative analysis of complex biologic lipid mixtures (1, 2), as well as to the quantitation of selected biochemical entities (3). Quantitation by TLC has been limited, for the most part,to the recovery of separated components from the chromatoplate, followed by an independent analysis using established analytic methods. Current te...

OBJECTIVES To investigate the feasibility of targeting ceramide metabolism to enhance chemotherapy cytotoxicity in prostate cancer. Discovering new targets for cancer treatment is an important endeavor, especially in prostate malignancies, which often revert to hormone- and chemotherapy-refractory disease states. METHODS Ceramide metabolism was measured in human prostate cancer cell lines usi...
متن کاملAnalysis of brain lipids by high performance thin-layer chromatography and densitometry.
We have devised a high performance thin-layer chromatography (HPTLC) densitometry method to resolve the major lipid classes of brain tissue. We used DEAE-Sephadex column chromatography to separate the total lipid into neutral and acidic lipid fractions. The lipid fractions were then spotted on separate HPTLC plates and chromatographed in one dimension using two solvent systems. Quantitation was...
متن کاملA method for microdetermination of major neutral lipids and an application of the procedure to tissue lipids.
Cholesterol, cholesteryl esters, triglycerides and fatty acids as major neutral lipids and phospholipids were examined in quantitative analysis. The method consisted of three steps: (1) separation of lipids by one-dimensional thin-layer chromatography on silica gel plates; (2) elution of neutral lipids from scraped silica gel with chloroform-methanol (4:1); and (3) colorimetric determination of...
